اثر مکمل خوراکی لوابون بر کارایی رشد و بیان ژن های مرتبط با رشد، سیستم ایمنی و آنتی اکسیدانی در ماهی کپور معمولی (Cyprinus carpio)

چکیده

این تحقیق به منظور تعیین اثر لوابون (مخمر اتولیز شده Saccharomyces cerevisiae) بر کارایی رشد، بیان برخی ژن‌های مرتبط با رشد (IGF-I)، سیستم ایمنی (LYZ و TNF-1α) و آنتی‌اکسیدانی (SOD و CAT) در بافت روده و کبد ماهی کپورمعمولی انجام شد. تعداد 240 قطعه ماهی با میانگین وزنی 0/67± 14/53 گرم در 12 حوضچه فایبرگلاس 250 لیتری در قالب یک طرح کاملاً تصادفی شامل 4 تیمار و 3 تکرار توزیع شدند. ماهیان بطور روزانه به میزان 3 درصد وزن بدن با جیره‌های غذایی حاوی سطوح 0، 0/3، 0/6 و 0/9 % لوابون به ازای هر کیلوگرم جیره به مدت 8 هفته غذادهی شدند (به ترتیب تیمار شاهد، اول، دوم و سوم). در پایان آزمایش ماهیان وزن‌گیری و از بافت کبد و روده نمونه‌برداری انجام شد. RNA از نمونه‌های بافتی استخراج و cDNA سنتز و با آغازگرهای اختصاصی توسط Real time PCR میزان بیان ژن‌های مرتبط اندازه‌گیری شد. نتایج نشان داد در عملکرد رشد بین تیمارهای آزمایشی با یکدیگر اختلاف معنی‌داری وجود ندارد. میزان بیان ژن لیزوزیم (LYZ) در بافت روده در بین تیمارهای آزمایشی و تیمار شاهد اختلاف معنی‌داری نشان نداد. میزان بیان ژنهای TNF-1α، IGF-I،SOD و CAT در بافت روده و کبد بین تیمارهای آزمایشی و تیمار شاهد اختلاف معنی‌داری نشان داد و بیشترین میزان بیان ژن در تیمار آزمایشی دوم مشاهده گردید (0/05 > P). بنابراین سطوح مختلف لوابون بر عملکرد رشد ماهی کپورمعمولی اثرمعنی‌داری نداشت و استفاده از سطح0/6 درصد در افزایش بیان ژن‌های مرتبط با سیستم ایمنی و آنزیمهای آنتی‌اکسیدانی در ماهی کپورمعمولی موثر می باشد.

The aim of this study was to investigate the effect of Levabon® (autolyzed yeast, Saccharomyces cerevisiae) on growth efficiency, the expression of growth (IGF-I), immune (TNF-1α, LYZ) and antioxidant (CAT, SOD) related genes in the intestinal and liver tissue of common carp (Cyprinus carpio). The fish (n=240; 14.53 ± 0.67 g) were fed with 0.0, 0.3, 0.6, and 0.9% of Levabon® in 12 tanks (250 L) with three replicates (each with 20 fish) for 8 weeks. At end of experiment, fish were weighted and liver and intestinal tissue were collected. RNA was extracted from tissue samples and cDNA was synthesized and the expression of related genes was measured by real time PCR with specific primers. The results of measured experimental indices revealed that there is no significant difference in the growth performance and intestinal lysozyme (LYZ) gene expression among experimental treatments. The expression of TNF-1α, IGF-I, SOD and CAT genes in the intestinal and liver tissues showed a significant difference between the experimental treatments and the control, and the highest gene expression was observed in the 0.6% experimental treatment (P<0.05). Overall, these results suggested that inclusion of Levabon in common carp diet had no significant effect on growth performance but can beneficially effect on immune and antioxidant system.
