اثرات به‌کارگیری پروبیـوتیک لاکتـوباسیلوس پلانتاروم(Lactobacillus plantarum) بر هورمون جنسی استرادیول و بافت گنادی در جنس ماده ماهی گورخری (Danio rerio)

چکیده

در این تحقیق تأثیر سطوح مختلف پروبیوتیک لاکتوباسیلوس پلانتاروم در جیره غذایی بر هورمون های جنسی استرادیول و بافت گنادی ماهی گورخری بررسی شد. تعداد 600 قطعه بچه‌ماهی بامیانگین وزنی01/0±15/0 ‌گرم در 4 تیمار و 3 تکرار با جیره‌ی غذایی پایه همراه با سه سطح پروبیوتیک لاکتوباسیلوس پلانتاروم (CFU/gr 105، 106 و107) به مدت سه ماه تغذیه شدند. در پایان دوره و مشخص شدن جنسیت، جنس نر از آزمایش خارج و جنس ماده به مدت 1 ماه به‌ وسیله همان جیره‌های تغذیه ‌شدند. در انتهای دوره از کل بدن ماهی استفاده گردید و پس از تهیه سرم، به روش RIA میزان هورمون‌های استروئیدی اندازه گیری گردید. سـپس بخشی از بافت گناد نمونه برداری و میکروسکوپ نوری بررسی شد. نتایج حاصل از این پژوهش نشان‌دهنده افزایش این هورمون در زمان ویتلوژنز می‌باشد که در تیمار تغذیه‌شده با CFU/gr106 لاکتوباسیلوس پلانتاروم افزایش معنی‌داری نسبت به بقیه تیمارها نشان می-دهد(P<0.05). ولی تیمار تغذیه‌شده با CFU/gr107 لاکتوباسیلوس پلانتاروم روند کاهشی داشته است. در مقاطع بافتی در تیمار شاهد بیشتر تخمک‌ها به وضوح در مرحله‌ی دوم و سوم رسیدگی جنسی قرار دارند. در تیمار تغذیه‌شده با CFU/gr106 لاکتوباسیلوسپلانتاروم بخش عمده تخمک‌ها در مرحله سوم و چهارم و حتی انتهای مرحله چهارم بلوغ بودند. که نسبت به تیمارهای تغذیه‌شده با CFU/gr 105و CFU/gr107 لاکتوباسیلوس پلانتاروم این مراحل بیشتر مشاهده می‌شود. نتایج مطالعه حاضرنشان می‌دهد که استفاده از پروبیوتیک‌ها لاکتـوباسیلوس پلانتاروم در مقدار موثر می‌تواند به‌عنوان یک راهبرد جدید در بهبود عملکرد تولیدمثلی مورد توجه قرار گیرد.

چکیده [English]

The objective of this study was to investigate the effect of different levels of Lactobacillus plantarum probiotic on estradiol hormone and gonad tissue in zebrafish (Danio rerio). So, six hundred healthy zebrafish (0.15+0.01 gr) were randomly distributed in 12 glass aquariums and fed experimental diets contain different levels of L. plantarum (0%, 105, 106 and 107CFU/gr ) for 3 months . After this period and sex determination, males were seprated and females fed probiotic for 1 month more. At the end of the experiment, the whole body of the fish was used to measure the hormone, the amount of esteroid hormone was measured by RIA method. Also of the histological point of view were studied by light microscope. The results of this study showed an increase in the amount of estradiol hormone during vitellogenesis in fish fed 106 CFU/gr L. plantarum which had a significant difference with other treatments (P <0.05). However, 107 CFU/g fed L. plantarum treatment had a decreasing trend. In tissue sections, the most of the eggs in control were clearly in the second and third stages of sexual maturation while in the 106 CFU/gr fed treatment, most of the eggs were in the third and fourth stages and even at the end of the fourth stage of maturation.These stages were more common than treatments fed 105 CFU/g and 107 CFU/g L. plantarum. The results of the present study indicated that the use of probiotics L. pentarum in the effective amounts can be considered as a new strategy to improve reproductive performance.
