تاثیر غلظت‌های تحت کشنده نیترات نقره (AgNO3) بر شاخص‌های بیوشیمیایی و بیان ژن سیتوکروم P450 در ماهی کپور معمولی (Cyprinus carpio)

چکیده

هدف از این تحقیق بررسی اثرات تحت کشنده نیترات نقره بر بیان ژن P450 و برخی شاخص‌های بیوشیمیایی(در ماهی کپور معمولی می‌باشد بچه‌ ماهیان با میانگین وزنی33/0± 7 گرم تهیه و در3 تیمار و 1 گروه شاهد در 3 تکرارتقسیم بندی شدند. ماهیان در معرض غلظت‌های موثر ppb23/0، 68/0و13/1 به مدت 14 روز قرار گرفتند. در پایان دوره، خونگیری و نمونه‌گیری از بافت کبد و آبشش جهت سنجش شاخص‌های بیوشیمیایی و ارزیابی بیان ژن P450 صورت گرفت. نتایج نشان داد که بیان ژن P450 در آبشش در تیمار 3 به صورت معنی‌داری نسبت به شاهد افزایش یافت (05/0P<). همچنین کم‌ترین میزان بیان ژن در آبشش در تیمار 1 مشاهده شد. نتایج نهایی در ارتباط با قرارگیری در معرض نیترات نقره نشان داد که با افزایش مدت زمان و غلظت سم نیترات نقره اثرات قابل توجهی بر شاخص‌های سرمی ماهی کپور معمولی داشت. پروتئین کل در تیمارهای 2 و 3 به طور قابل توجهی در مقایسه با گروه شاهد کاهش یافت (05/0> P). سطح ایمنوگلوبولین کل با افزایش غلظت سم افزایش یافت، اما این افزایش معنی‌دار نبود (05/0> P). سطح آلبومین در تیمار 3 به طور معنی‌داری کمتر از گروه شاهد بود (05/0> P) اما در سایر تیمارها تفاوت معنی‌داری با گروه شاهد مشاهده نشد همچنین میزان بیان ژن P450 تحت تاثیر سم در کبد و آبشش در روز 14 بالاترین میزان بیان را نشان داد. بنابرابن می‌توان چنین استنباط نمود نیترات نقره توانست آثار مخربی را بر ماهی کپور معمولی که یکی از مهم‌ترین ذخایر دریای خزر است، اعمال کند.

چکیده [English]

The purpose of this study was to investigate the effect of sub-lethal concentrations of silver nitrate (AgNO3) on some biochemical indices and P450 gene expression of common carp. For this purpose, Common carp with a weighing average of 7 ±0.33 g were divided into 3 treatments (T1: 0.23, T2: 0.68 and T3: 1.13) and 1 control group, each containing 3 replicates, exposed to the effective concentrations of 0.23, 0/68 and 1/13 ppb for 14 day. At the end of the study, blood, gills and liver samples were collected for evaluate of some biochemical indices (total protein, Immunoglobulin, albumin) and P450 gene expression. The results showed that P450 gene expression in the gills of treatment 3 was significantly increased compared to the control group (P <0.05). The final results of this study in connection with exposure to silver nitrate toxin showed that with increasing duration and concentration of silver nitrate toxin has significant effects on biochemical indices of common carp and in most experiments reduces these indices. Total protein in the 2nd and 3rd groups significantly decreased compared to the control group (P <0.05). Total immunoglobulin levels increased with increasing concentrations of toxin, but this increase was not significant (P> 0.05). Albumin level in treatment 3 was significantly lower than control group (P <0.05) but in other treatments no significant difference was observed with the control group. Also, the expression of P450 gene under the influence of toxin in the liver and gills on day 14 showed the highest expression. Therefore, it can be concluded that silver nitrate can have destructive effects on common carp, which is one of the most important resource of the Caspian Sea.
