This research was conducted to evaluate the effects of using the Saccharomyces cerevisiae probiotic on growth indices and the expression of growth-related genes (GH and IGF-1) in koi fish (Cyprinus carpio L.). To achieve this goal, 132 Koi carp fry, weighing approximately 20 ± 1.6 g, were obtained from a private sector hatchery. After a period of acclimatization to breeding environment conditions and hand-feeding, they were subjected to experimental treatments for 8 weeks, including the Saccharomyces cerevisiae probiotic Vivacell in 4 groups (0, 0.1, 0.5, 1%) with three replicates. At the end of the period, samples were taken from the treatments in a completely randomized manner, and growth indices were examined. To measure gene expression, brain and liver tissue samples were taken, RNA was extracted, and cDNA was synthesized using a synthesis kit. Gene expression was performed using primers for the (GH and IGF-1) genes and the beta-actin gene in Real-Time PCR.
Based on the results obtained, in Koi carp fry, different levels of the probiotic showed a significant effect on final weight and final body length, feed conversion ratio, specific growth rate, feed intake, and weight gain, as well as the number of red and white blood cells, hemoglobin concentration, mean corpuscular hemoglobin concentration (MCHC), and mean corpuscular hemoglobin (MCH) (p < 0.05). Evaluation of IGF-1 and GH gene expression in the 0.5 and 1% treatments showed a significant increase compared to the control group and the 0.1% treatment (p < 0.05).
The results of this study indicate the beneficial effects of the Saccharomyces cerevisiae Vivacell probiotic on growth indices and the expression of growth-related genes (GH and IGF-1) in Koi carp fry.
