In this study, the genetic structure of the Monkey goby population (Neogobius fluviatilis) of the Caspian Sea was investigated using microsatellite method. For this purpose, 90 samples were collected from Gomishan wetland, Gorgan River and Gorgan Bay in Golestan province. Genomic DNA was extracted using phenol-chloroform method. Polymerase chain reaction (PCR) was performed using seven pairs of microsatellite primers and the product was electrophoresed on 8% polyacrylamide gel and then stained with silver nitrate. All applied loci showed polymorphism. The results showed that Monkey goby has proper allelic diversity (mean number of alleles: 2.466) and heterozygosity (average heterozygosity observed: 0.44) in the studied areas. Of the 21 loci-population locus tests, only 1 were in the Hardy-Weinberg equilibrium. Fst value as an indicator of genetic differentiation was 2.5. In this regard, the relatively high gene flow (Nm:4.491) could be considered as an important factor in the low genetic differentiation between the samples of the studied area. The observed gene flow is probably related to factors such as contamination, floods, and the species life history. The results of molecular analysis of variance also showed that most of the observed diversity is within populations. The highest similarity was observed between the samples of Gorgan River and Gorgan Bay.
