ارزیابی روش سطح پاسخ بر تغییرات نقطه ذوب و دمای ژله‌ای کاراگینانflagelliformis Hypnea

چکیده

بیوپلیمر دریایی کاراگینان از دیواره سلولی کارژینوفیت‌های قرمز استخراج می گردد. در این پژوهش تاثیر شرایط مختلف استخراج بر نقطه ذوب و دمای تشکیل ژل کاراگینان استحصالی از کارژینوفیت flagelliformis Hypneaبا روش رویه سطح پاسخ (RSM) مورد بررسی قرار گرفت. بازه متغیرهای ورودی شامل غلظت قلیا (2-25/0 درصد w/v)، دمای استخراج (90-70 درجه سانتیگراد) و زمان استخراج (60-240 دقیقه) در 5 سطح بودند. مطابق با نتایج، تمامی متغیرهای ورودی بر نقطه ذوب معنادار بودند (05/0p<). در صورتی که غلظت قلیا تنها متغیر خطی ورودی بر دمای تشکیل ژل کاراگینان بود. تعیین نقطه بهینه برای نقطه ذوب و دمای تشکیل ژل با مدل چندجمله‌ای محاسبه گردید. مقادیر بهینه متغیرهای مستقل با استفاده از روش تابع مطلوب برای غلظت قلیا، دما و زمان استخراج به ترتیب 54/1 درصد، 85 درجه سانتیگراد و 203 دقیقه بودند. آزمایشات تاییدی برای نقطه ذوب و دمای تشکیل ژل به ترتیب 91/0±81/63 درجه سانتیگراد و 62/0±29/37 درجه سانتیگراد بدست آمدند. تطابق مقادیر واقعی با مقادیر پیشنهادی حاکی از توانمندی مدل بود (05/0p>).

چکیده [English]

Carrageenan is a marine biopolymer that can be extracted from cell wall of some red carrageenophyte. In this present, response surface methodology was used to investigate the procedure of carrageenan melting point and gelling temperature from algae of Hypnea flagelliformis. Independent variables, alkalinity concentration (0/25-2%), extraction temperature (70-90°C) and extraction time (60-240 minutes) were applied in five levels. all
Base on the result, effects of all independent variables on melting point were significant (p<0.05), whereas alkalinity concentration had significant effect on gelling temperature (p>0.05). Polynomial models developed for the melting point and gelling temperature indicated that optimum conditions. Optimum data for independent variables were alkalinity concentration 1.54%, temperature 85 °C and time 203 minutes.
Confirmatory test was earned as follows melting point 63.8 ±0.91 °C and gelling temperature 37.29 ±0 /62 °C. The experimental data and model forecasting were similar (P> 0.05).
