کاربرد نشانگرهای ریزماهواره و ژنوم میتوکندریایی در بررسی‌های ژنتیک جمعیت ماهیان

چکیده

حفاظت از منابع ژنتیکی در جوامع ماهیان، در درجه اول نیازمند آگاهی از میزان ذخایر توارثی و تنوع‌ژنتیکی بین افراد یک گونه و نیز نیازمند حفظ و نگهداری منابع ژنتیکی در مراکز تکثیر و جوامع وحشی یا طبیعی است. در صورت عدم مدیریت صحیح در برنامه‌های تکثیر و رهاسازی ماهیان احتمال بروز خطراتی از جمله از دست رفتن تنوع ژنتیکی درون جمعیتی، بین جمعیتی و در نهایت انقراض وجود دارد. از اهداف کلی تحقیقات ژنتیک جمعیت، تشخیص وسعت تنوع-ژنتیکی داخل گونه‌ها و محاسبه این تنوع می‌باشد و در ژنتیک جمعیت ترکیب ژنتیکی و تکامل جوامع مختلف مطالعه می-شود. جهت تخمین تنوع‌ژنتیکی جمعیت‌ها از نشانگرهای گوناگونی استفاده می‌شود. نشانگرهای مولکولی مبتنی بر DNA در سال‌های اخیر جایگزین مناسبی برای نشانگرهای قدیمی مورفولوژیکی و سیتوژنتیکی محسوب می‌شوند. نشانگرهای ژنوم میتوکندریایی mtDNA از گروه نشانگرهای غیرمبتنی بر PCR و نشانگرهای ریزماهواره (میکروستلایت) از گروه نشانگرهای مبتنی بر PCR، به دلیل مزایای متعدد هر دو نوع نشانگر که تنوع و هتروزیگوتی را به خوبی نشان می‌دهند، کاربرد فراوانی در مطالعات ژنتیک جمعیت دارند. از میان نشانگرهای مولکولی که در سطح mtDNA استفاده می‌شوند، ژن 16SrRNA به کاربرد فراوانی در بررسی‌های فیلوژنتیک ماهیان دارد. همچنین میکروستلایت‌ها در بررسی تنوع‌ژنتیکی درون و میان جمعیت‌های یک گونه بسیار ارزشمند و کارآمد می‌باشند.

The conservation of genetic resources in aquatic animals needs awareness about genetic reserves and genetic diversity among one species' individuals and also needs the conservation of genetic resources in the hatchery or wild and natural communities. It will lead to some threats such as inter- and intra-population genetic diversity loss and species extinction if propagation and sea ranching program being conducted improperly. Determination and calculation of inter- and intra-assay genetic diversity are of general goals of population dynamic studies and population genetic studies the genetic cross and population evolution. Different markers are used to estimate populations' genetic diversity. In recent years, DNA-based molecular markers have been considered as suitable alternatives for old morphologic and cytogenetic ones. Mitochondrial genomic markers mtDNA belonging to non-PCR-based markers and microsatellite markers belonging to PCR-based markers show diversity and heterozygocity suitably, thus have wide use in population genetic studies. From the molecular markers at mtDNA level, 16SrRNA is widely used in fish phylogenetic studies. On the other hand, microsatellites markers are very efficient and worthy in inter- and intra-population genetic diversity studies.
