تعیین سمیت کشنده (LC5096h) نانوذرات نقره روی دافنی ماگنای بالغ، Daphnia magna

چکیده

استفاده از نانو ذرات نقره (با اندازه کمتر از 100 نانومتر) به‌ویژه در حوزه سلامتی به‌صورت گسترده‌ای توسعه ‌یافته است. دافنی ماگنا (Daphnia magna) به دلیل سادگی تکثیر و حساسیت بالابه تغییرات محیطی دارای کاربرد گسترده ای در مطالعات مربوط به آلودگی بو سازگان آبی می باشد. هدف از تحقیق حاضر، تعیین سمیت کشندگی (LC50 96h) نانو ذرات نقره بر دافنی ماگنا بالغ بود. نمونه های دافنی ماگنا از استخرهای پرورش ماهی در گنبد تهیه و در آزمایشگاه در شرایط کنترل‌شده (دمای 1±20 درجه سانتی‌گراد و 8:16 ساعت روشنایی/تاریکی اکسیژن محلول معادل 80/8 میلی‌گرم در لیتر، پی اچ معادل 8 ، سختی کل معادل 293 میلی‌گرم در لیتر، شوری معادل 86/1 قسمت در هزار) قرار گرفت. سپس از یک دافنی به منظور خالص سازی، 3 نسل متوالی تهیه شد، تست سمیت حاد (LC50) در 4 غلظت مختلف (25/0، 5/0، 1 و 2 قسمت در میلیون) به همراه شاهد در 3 تکرار انجام گردید و نتایج مرگ‌ومیر در طی زمان‌های 24، 48، 72 و 96 ساعت اندازه‌گیری گردید. در این پژوهش میزان 48h LC50 نانو ذرات نقره بر دافنی ماگنا 416/0 قسمت در میلیون بدست آمد. به‌طورکلی می‌توان نتیجه گرفت که نانو ذرات نقره سمیت بالایی برای دافنی ماگنا داشته و این سمیت با افزایش غلظت و همچنین با افزایش مدت مجاورت افزایش معنی‌داری نشان می دهد.

The use of Silver nanoparticles (with a size less than 100 nm), especially in the field of health is widely developed. Daphnia has wide application in pollution studies of aquatic ecosystems due to the simplicity the top to reproduce and sensitive to environmental. The aim of this study was to determine the lethal toxicity (LC50 96h) silver nanoparticles on Daphnia magna adult. Samples of D.magna were prepared from fish farms in the Gonbad region and reared in the laboratory under controlled conditions (temperature 20±1°C; 16h:8h light:dark; dissolved oxygen, 8.80mg/liter; pH 8; total hardness, 293 mg/liter; salinity to 1.86 ppt). Then in order to purification of a daphnia, three consecutive generations were prepared. Acute toxicity test (LC50) was achieved at four different concentrations (0.25, 0.5, 1 and 2 parts per million) along with control and three replications and mortality in periods of 24, 48, 72 and 96 hours were measured. In this study, L C5048h for silver nanoparticles was estimated 0.416 part per million (ppm) on D.magna. Overall, it seems that silver nanoparticles show high toxicity for D.magna and significant increase by increasing the concentration and exposure time.
